肝素酶主要從一些利用肝素為碳源的細(xì)菌中分離得到,*初來源于肝素黃桿菌(Flavobacterium heparinum或Pedobacter heparinus)。此外,在許多微生物中也發(fā)現(xiàn)了肝素酶的存在,如棒桿菌(Corynebacterium sp.)、鞘胺醇桿菌(Sphingobacterium sp.)、枯草芽孢桿菌(Bacillus subtilis)、環(huán)狀芽孢桿菌(Bacillus circulans)、解肝素?cái)M桿菌(Prevotella heparinolytica)以及糞便擬桿菌(Bacteroides stercoris HJ-15)等。但至今肝素黃桿菌仍然是商品肝素酶的**來源。已從肝素黃桿菌中分離純化出的肝素酶有三種,分別為肝素酶I、II、III,這三種酶的酶學(xué)特性各不相同。在肝素黃桿菌純化出的三種肝素酶中,只有肝素酶II的結(jié)構(gòu)被解析,它的底物特異性也*差;肝素酶I則*適用于低分子量肝素的制備。
