The new Badrilla SiteCounterTM S-Palmitoylated protein kit is a quick and easy way to quantify the extent of S-palmitoylation of your protein of interest. Using a straightforward mass tag approach, modification of each cysteine residue results in an incremental increase in the mass of the protein. The number of mobility shifts (as detected by western blot analysis) is observed as a ladder of protein bands, each ‘step’ in the ladder representing an additional S-palmitoylated cysteine residue.

There have been many recent advances in methodologies to detect the only reversible lipidation reaction of proteins, S-palmitoylation. This fatty acyl post translational modification has gained great interest in the research community as it is increasingly recognised as important in both health and disease. In this article I will briefly examine the development of the acyl exchange methodologies used by researchers to measure S-palmitoylation. I will focus in particular on the recently published Acyl PEG Exchange (APE) /Acyl PEGyl Gel Shift (APEGS) assay, paying particular attention to the advantages it offers scientists interested in exploring palmitoylation.
