Reaction Conditions 
2X Buffer UB
50mM Tris-acetate (pH 7.6 at 30°C), 20mM Mg-acetate, 200mM K-acetate, 14mM 2-mercaptoethanol, and 100μg/ml BSA. Incubate at 37°C.

Storage Buffer
10mM Tris-HCl (pH 7.5), 200mM KCl, 0.1mM EDTA, 1mM DTT, 100μg/ml BSA and 50% glycerol. Store at -20°C

Thermal Inactivation
65°C for 20 minutes.

Ligation / Recutting Assay 
After 2-fold overdigestion with BssMI, 90% of the DNA fragments can be ligated and recut.

Overdigestion Assay
